Summary:Optimization of small scale lipid extraction method for freshwater fish (Barbodes banksi) and marine fish (Polydactylus sextarius) muscles are conducted based on Folch (1957) lipid extraction procedure. The optimization process was conducted by comparing different homogenization methods, extraction solvent ratios, and sample weight to solvent ratios to achieved high percentage of lipid recovery. On the first optimization, three methods of tissue homogenization (glass beads, Mortar and pestle, and mincing by dissecting blade) were done and glass beads was found to be optimal method for both freshwater fish and marine fish muscle tissue. Second optimization was done by using three different ratio of extraction solvent (v/v; 1: 2, 2: 1 and 2.5: 0.5). Solvent to solvent ratio of 2: 1 was best ratio for freshwater fish muscle whereas solvent to solvent ratio of 1: 2 was best for marine fish muscle. Third optimization was done by using three different ratio of sample weight to solvent (w/v; 1: 10, 1: 20 and 1: 40). The standard Folch (1957) sample weight to solvent ratio 1: 20 is efficient for freshwater fish muscle. However, for marine fish muscle it gives highest lipid recovery for sample weight to solvent ratio 1: 40. Optimal small scale lipid extraction procedures for two different nature of tissue which is for freshwater fish and marine fish muscles were proposed. Further study on lipid composition is recommended to determine the effect of the different extraction procedures on recovery of various lipid contents.
