There is a wide range of Rasbora that had been discovered particularly in Southeast Asia, but the cytogenetics characterization of this genus still remains limited. With the purpose of understanding the Rasbora spp. chromosomal configuration, cytogenetic analysis had been conducted on this genus. In this research, RPMI 1640 medium was used for cell culturing and this media contained 1 % Pen Strep and 10% FBS to aid in the optimization of media. In the preparation of chromosome, three different organs were isolated to establish the cell lines, followed by sufficient sterilization in order to avoid any contamination. Next 0.1 % of colcemid was applied to the cells in order to inhibit the spindle fibres fonnation, thus arresting the cells in the metaphase stage. Once arrested, cell fixation was perfonned using the Camoy's fixative (M:AC). The cells were dropped on several slides and were then stained with Giemsa stain to study the banding pattern on the chromosome. However, in this study, the chromosome obtained after giemsa staining appeared to be clumping and overlapping, hence only pink patches was observed. Further studies on karyotyping method are needed to obtained better and clearer result.
