Summary:Knowledge of genetic diversity in rice (Oryza sativa L.) germplasm has significant impact in the improvement program. The study is to determine the level of genetic variation and reveal the genetic diversity among selected Sarawak traditional rice using RAPD technique. Two random primers (OPA13 and OPAI6) were used to amplify DNA segments extracted. The results indicated that 17 alleles (94.5%) were polymorphic and 1 allele (5.5%) was monomorphic. The highest genetic similarity coefficient was 0.9655 between Padi Avong and Padi Sawah and lowest was 0.5217 between Adan Merah and Bario. A dendogram based on similarity index using NTSYSpc among 10 Sarawak traditional rice accessions group into three major groups. Findings from this study suggested that the RAPD molecular techniques are effective towards the identification of rice genetic germplasm in Malaysia.
