Summary: | Bromelain is an aqueous extract, obtained mainly from stems of young pineapples although it is present in all parts of the plant. It contains proteases and non-protease compounds that contribute to its medicinal properties, such as anti-thrombotic, anti-inflammatory, anti-proliferative and anti-cancer. Previous studies have proved the anti-cancer properties of bromelain on cancers of the lung, brain, colorectum, skin and breast, but no studies have been reported on nose and throat cancer; nasopharyngeal carcinoma (NPC). Therefore, the anti-proliferative effects of bromelain on a panel of
human nasopharyngeal carcinoma cell lines HK 1, HONE -1, SUNE -1, TW 01, and TW04, were investigated byutilizing the tetrazolium salt,3-(4,5dimethylthiazol-2-yl)
-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H- tetrazolium, MTS Assay Kit.
In addition, effect of bromelain on cell apoptosis was examined based on mitochondria membrane potential, by Mitochondria Staining Kit. In this study, bromelain significantly suppressed cell proliferation of NPC cell lines in an increasing dosage manner, without radiation. Correspondingly, apoptotic bodies were detected by fluorescence microscopy, in treated cells, through mitochondria staining. These results confirmed that bromelain exerts cytotoxic effects on the cell growth and endogenous activities of NPC cell lines in vitro by
demonstrating its ability to suppress cell proliferation and induce apoptosis without radiation. The results also suggested possible bromelain-induced extracellular signalling pathways(NF-kB, MAPK /Erk)and caspase protein activation in NPC metastasis. |
