The intronic region of the human MYL9 (myosin light chain 9) gene was chosen in this study as in previous study there was no mutation found in the coding region when comparison was done between the normal and tumor tissue of colorectal. In particular, some introns play vital role in transcriptional control and splicing, so changes in these sequences will lead to clinical and circumstantial malfunction. The objective of this study is to isolate and clone the intron region between exon 1 and 2 of MYL-9 gene. Total genomic DNA extracted from the cultured cell line of nasopharyngeal and nasopharyngeal carcinoma was used as the starting material. Nasopharyngeal carcinoma (NPC), whose onset can be detected in the epithelial cells of the nasopharyngeal region, causes high incidence of fatality in patients mostly from South East Asia (Chang, 2006). Total of 3299 bp of intronic region between exon I and 2 of the MYL-9 gene was partially amplified by 1600 bp by polymerase chain reaction (PCR).
