Lipase is an enzyme which capable to be catalyst in the hydrolysis and the synthesis of esters formed from glycerol and long-chain fatty acids. Oil palm pressed fiber (OPPF) is an agricultural waste know as mesocarp fiber which recovered from pressed palm oil and based on dry weight basis, approximately it contains of 40 % cellulose, 24 % pentosan, 21 % lignin and 5 % ash. The production of extracellular lipase from OPPF using Aspergillus niger by solid state fermentation (SSF) was studied. The main target in this study is to produce high yield of lipase activity from OPPF based on media composition which is inducer, carbon and nitrogen sources. The optimized media composition consisted of OPPF of size 500 μm, 70 % of moisture content at 32 °C. In addition, it is desired to determine the effect of interaction between parameter, olive oil as inducer (0.5 % – 1.5 % v/v), sucrose as carbon source (0.5 % – 1.5 % w/v) and peptone as nitrogen source (0.1 % – 0.3 % w/v). Optimization of media composition was done by using Response Surface Methodology (RSM): Central Composite Design (CCD). In the optimization process, the design contains of a total of 17 experimental sample runs involving replications of central points. The optimization of lipase production by Aspergillus niger using OPPF as solid substrates shown a maximum production of about 0.3763 U/mL of lipase activity which obtained from experimental run which contains of 0.37 % (v/v) of olive oil, 0.20 % (w/v) of sucrose and 0.04 % (w/v) of peptone in the fermentation medium. 2013-08-26T09:13:32Z 2013-08-26T09:13:32Z 2012-06 Learning Object http://hdl.handle.net/123456789/27672 en Universiti Malaysia Perlis (UniMAP) School of Bioprocess Engineering
