Summary:This study is aim to determine production of cellulase production by using lignocellulosic waste from palm oil. The lignocellulosic waste used in this study is empty fruit branch of palm oil. It is pretreated with alkaline solution before undergoes fermentation process to produce cellulase. Pretreatment process able to break down the structure hard especially lignin to more simple substrate for Bacillus sp. consume. Bacillus subtilis IBRL A3 are selected as source of cellulase production bacteria, it give a better clear zone in congo red test. Extracted cellulase is filter by using filter paper to remove the substrate and centrifuge at 10000 rpm for 10 minutes to obtain the filtrate. Later filtrate is assay by using the DNS reagent and observes under spectrophotometer at 540 nm. There are three factor focused in production of cellulase, incubation time, temperature, inoculums size. Based on this study optimum of incubation time is at 72 hour which recorded as 7.76 U/g. While, optimum temperature to cellulase production is at 35°C recorded as 0.76 U/g. Besides that, optimum inoculums size found is 30% based on this study as recorded 9.72 U/g. Hence, production of cellulase optimum at 35°C, 30% inoculums size for 72 hour.
