Summary: | This study was conducted to determine the best sterilization method for Hevea brasiliensis and the concentration of hormonal regulation of plant combinations are best suited for the development and growth of rubber trees through tissue culture methods. The major problem faced in the study is bacterial contamination. It might due to contaminated explant used with bacteria and other microorganisms. Different sterilization methods were tested to overcome this problem. Based on this parameter, the best result was obtained with the combination of 80% (v/v) of ethanol for 20 seconds and 4% (v/v) NaOCI for 3 minutes (Method 2). Further study were carried out to investigate the effect of cytokinin on shoot initiation in three different concentration of BAP/Kn/2ip and constant NAA/IAA/2 BAP/Kn/2ip,4-D. Result shown that MS media supplemented with 0.5mg/L IAA + 1.0mg / 1 2ip give shoot after 2 weeks of culture. Further studies on Hevea brasiliensis should be carried out to determine the best conditions for shoots regeneration in Hevea brasiliensis. |
